TitrationAnalysis: a tool for high throughput binding kinetics data analysis for multiple label-free platforms

Label-free techniques including Surface Plasmon Resonance (SPR) and Biolayer Interferometry (BLI) are biophysical tools widely used to collect binding kinetics data of bimolecular interactions. To efficiently analyze SPR BLI data, we have built a new high throughput analysis tool named the TitrationAnalysis. It can be as package in Mathematica scripting environment ultilize non-linear curve-fitting module for its core function. This fit time course estimate association dissociation rate constants (ka kd respectively) determining apparent constant (KD) values. The fitting process is automatic, requires minimal knowledge on applied from multiple label-free platforms. We demonstrate that TitrationAnalysis optimal antibody-antigen acquired Biacore T200 (SPR), Carterra LSA (SPR imaging) ForteBio Octet Red384 ka, KD values derived using very closely matched results commercial software provided specifically these instruments. Additionally, generates user-directed customizable output readily downstream Data Quality Control associated with Good Clinical Laboratory Practice operations. With versatility source input options result output, offers investigators powerful alternative biomolecular interaction characterization.
